DESIGN OF GASTRORETENTIVE POLYMERIC LOW-DENSITY MICROBALLOONS OF MEBENDAZOLE USING RESPONSE SURFACE METHODOLOGY

Objective: The main objective of the present work is development polymeric low-density microballoons for prolonged gastroretentive delivery and optimization promising formulation by central composite design response surface methodology. Methods: Mebendazole-loaded were prepared emulsion solvent diffusion method using Eudragit S-100 hydroxypropyl methylcellulose as release controlling polymers. All formulations mebendazole-loaded showed buoyancy up to 8 h. Percentage (X1) in total amount polymer ratio (X2) was taken two independent variables. responses are evaluated study effect variables optimum chosen based on numerical graphical optimization. Results: optimized MBZ9 composed 100 mg mebendazole, 75% S-100, 25% HPMC with DCM: ETH 1:1. yield (81%), (86.4%), entrapment efficiency (82.01%), cumulative drug 12 h (79.99%). characterized differential scanning calorimetry, Fourier-transform infrared spectroscopy. It followed mixed order mechanism per R2=0.905 Higuchi model. Conclusion: Microballoons mebendazole produced X1 (750 mg), polymer, 1:1 X2 (10:10 ml) methodology successful enhanced controlled elicit anthelminthic gastrointestinal tract.
